According to Simon (2002) the silica gel is the most widely used absorbent and it is the stationary phase that is dominant in Thin layer chromatography, particles or molecules that are non-polar travel faster at the stationary phase and the molecules that are polar spend more time at the stationary phase and therefore this means that molecules that are non-polar travel larger distance on the TLC plate in comparison to molecules that are polar since the polar molecules stick to the polar stationary phase (gel silica plate). According to Mary (1996) the molecules or substances move or travel up the stationary phase by means of capillary action and the stationary phase is polar because of the -OH groups which are polar and are bound to the silica …show more content…
According to voet etl (2013) the amino acid lysine and glutamic acid are amino acids that are polar and their polarity comes their sides chains, and the amino acids glycine and leucine are none polar and therefore this means they will move longer distances on the TLC plate. The amino acid lysine and glutamic acids travelled shorter distances on the TLC plate and this is because this two amino acids are polar. Lysine migrated a distance of 9.5mm on average and 20 mm and their Rf values are 0,14 and the 0,30 and this Rf values are smaller compared to glycine and leucine, for glutamic acid it did not have any migration on the TLC and their Rf was 0. Leucine and glycine migrated larger distance on the TLC place and their Rf values are bigger compared to those of glutamic acid and lysine because the Rf value of leucine is 0,57 and Rf for glycine is 0,40. According to Lade etl (2014) the compound that have high polarity will have stronger affinity of binding to the polar stationary phase and therefore this means the polar molecules travel smaller distances on the TLC plate and non-polar molecules travel larger distances. The Rf values of the unknown mixture of mixture of amino acids are too small from this we can say that the unknown mixture of amino acids is polar since it travelled shorter distance on the TLC plate and their Rf values were 0,06 and 0,36. According to Harper and Davis (1979) the amino acids which have the Rf value that is closest to that of the unknown mixture is lysine which had the Rf value of 0,07 and the amino acid ornithine which had an Rf value of
