Objectives:

The purpose of this project is to identify two separate microbial organisms from a given list, which are to be inoculated from a mixed broth culture. Multiple tests will be performed in order to eliminate all organisms from the list that do not provide significant results for the identification of the unknown specimens.

Journal Entries:

April 17, 2018. Today we began the unknown project. I obtained the unknown broth tube #15. I used two Tryptic Soy Agar (TSA) plates in order to make a streak plate. The streak plate is used to separate the two different bacterial organisms that were present in the broth. The plates were placed in the incubator at 37C and in 48 hours there should be isolated colonies on the plates.

April 19, 2018. I obtained my TSA plate from the incubator and determined that there were two separate species present due to the size of the colonies. One species had very small colonies and the other species had larger colonies. I inoculated each different species with the aseptic loop technique and made two separate gram stains to determine the gram reaction and shape of the two separate species. I found that the bacteria that formed the smaller colonies are gram negative bacilli, and the bacteria that formed the larger colonies are

I started with the gram negative bacillus by performing the nitrate reduction test and the urease test. I chose these two tests because I knew that a positive urease test would indicate that the gram negative bacillus was P. rettgeri, but this test came back negative after I performed two just to make sure that the results were accurate. Also, the nitrate reduction test could have indicated that the gram negative bacillus was P. fluorescens, but the test result was positive after the reagents were added. Now I knew that I could rule out P. rettgeri and P. fluorescens for the gram negative

Gram Negative Unknown Lab Report #20 BIOL 3444-008 Giang Nguyen TA: Malini Kotak 11/05/2014 Abstract The experiment is conducted in order to identify a randomly given unknown gram-negative microorganism. By comparative analysis of biochemical tests that were performed throughout the semester to find out its characteristics and determine the identification of bacterium out of four potential unknown was given. To figure out the characteristics, T-streak was performed to isolate the sample from broth to a Tryptic Soy Agar plate, along with an additional slant in case something wrong with the original plate that can possibly affect the result.…

Following Gram staining and microscopic examination, Unknown 15 was identified to be a Gram positive coccoid. Based on these results, all of the Gram negative genera were eliminated including: Enterobacter, Citrobacter, Klebsiella, Escherichia, Pseudomonas, Serratia, Alcaligenes, Neisseria, Proteus, Salmonella, Shigella, Erwinia, Veillonella, and Flavobacterium (5). Bacillus, Lactobacillus, Listeria, Kurthia, and Arthrobacter were eliminated because they did not match the coccoid shape of Unknown 15 (5). The catalase test was the next designated test in the identification of Unknown 15. Unknown 15 tested negative for the enzyme catalase as it did not form gas bubbles after the addition of hydrogen peroxide to the sample.…

rosea or M. luteus. I conduct a Nitrate test to differentiate between nitrate and nitrite, in which nitrite can be further metabolized. My bacteria had a positive reaction with the A and B reagents by turning red to indicate nitrites presence. Therefore, I go on to perform a Gelatin Hydrolysis test which was negative, but may have had unreliable results due to the bacteria not having enough time to grow.…

During the phase and process of the bacterial unknown project, vast amounts of test were performed in order to determine an unknown species of bacteria. Within the many procedures of identifying the unknown bacteria, I was first given a tube with the unknown bacteria, from which later on I started subcultures by streaking bacteria from the original tube onto a tryptic soy agar (TSA) slant. Throughout the process of identifying the unknown, new subcultures were produced in order to use the newest bacteria to conduct the tests. Thus, this process allowed the unknown bacteria to grow and maintain. Before a series of test were conducted, first a gram stain test was performed in order to determine the unknown’s gram reaction and its cell morphology.…

A sample of the gram-positive cocci colony was taken out which then was transferred into a Urea broth to test for Urease. This was incubated at 37 degrees Celsius for 48 hours. The ending result was no color change, which was a negative reaction in the Urea broth. The results eliminated Staphylococcus epidermidis from being the bacteria in the specimen. This only left two candidates that were either Staphylococcus aureus or Enterococcus…

One test that was run to help in differentiating the bacteria is the motility test. The motility test is preformed by taking soft agar, and heavily inoculating it an inoculating needle. Introduction : Enterotube II is used to identify multiple tests in one tube. twelve different conventional media plus a self-contained inoculating wire. The twelve test are for glucose and gas, then lysine, ornithine, H2S and indole, adonitol, lactose, arabinose, sorbitol, dulcitol, phenylalanine (amino acid), urea, and citrate (No VP test require). These medias performs 15 different biochemical tests that will determine the genus and the species of gram negative bacteria. This test will help identify the 5digit number that will distinguish the organism using 5-digit interpretation code book.…

Once the Gram stains were complete, each sample was looked upon under a microscope as seen in figures 3 and 4. Bacteria A was found to be Gram-negative (fig. 3) due to displaying a predominately red color. Bacteria B was found to be Gram-positive (fig. 4) due to displaying a dominate blue color. Bacteria B was also noticeably a rod, or bacillus, shape which helped to further narrow down the possibilities to two bacteria: Bacillus cereus, and Bacillus…

The lab instructor issued out a test tube labeled with letter ‘I’, which consisted of two unknown bacteria, Gram-positive or Gram-negative that were streaked from a pure culture. Sterile techniques were followed while performing precise instructions as stated in the referenced Laboratory Manual. Example 1: The first procedure performed was done by isolating a pure culture from the mixture onto a solid Trypticase soy agar (TSA) media. Sterile technique was done by flaming the loop until it turned red to ensure that there were no current bacteria on the loop avoiding contamination followed by rapidly flaming the neck of the test tube to prevent the entry and contamination of unwanted microbes.…

A given test or media may fall into multiple categories. Selective and differential tests are generally less effective at species identification, and consequently, are normally only capable of presumptive identification (3). This lab uses different types of media that are both selective and differential. Mannitol Salt agar (MSA), which contains high levels of sodium chloride kills most bacteria that are not members of the halotolerant Staphylococcus genus. MSA also contains mannitol, which allows differential identification because an indicator detects acid produced by the fermentation of mannitol (4).…
