As described in the central dogma of biology, information from a gene can be used to build a protein in a two-step process. Transcription is considered to be the first step of gene expression. It is known to be the process of making a copy of genetic information stored in a DNA strand into a complementary strand of messenger RNA (mRNA). The process of eukaryotic transcription can be separated into three phases, initiation, elongation and termination. This complex process involves various cell signaling methods as well as the activity of many enzymes. Genes are segments of DNA nucleotides located on a chromosome that contains the codes for the construction of individual proteins. They are found in the nucleus of cells where transcription first takes place. The main enzyme that is involved in aiding the start of eukaryotic transcription is RNA polymerase II. With the help of transcription factors, it builds a single RNA strand in the 5’ to 3’ direction, complementing DNA’s template strand – the mRNA transcript. Transcription of a gene takes place in three different stages. Initiation of transcription requires a series of transcription factors as a host to assemble RNA polymerase II on a promoter. A transcription factor can recognize and bind to a sequence called a TATA box on the DNA that is found upstream at the eukaryotic promoter start site.
