What is an endospore and what does it do? |
An endospore is a dormant form of the bacteria that allows it to survive poor environmental conditions such as heat and chemicals. |
What is an endospore membrane made of and what is its function |
It has a tough outer covering made of the protein keratin which is the reason why it is resistant against heat, chemicals, and staining |
What is a primary stain in endospore staining and how is it applied |
Malachite green. Which is applied through Steam |
Malachite screen is a water soluble stain. What does this mean according to cell staining |
Vegetative cells and Spore mother cells can be decolorized with water while the endospores retain the malachite green and the rest ( vegetative cells and Spore mother cells) are counterstained with saffranin |
Where can spores be located inside of a cell |
In the middle of the cell- (Central) at the end of the cell- (terminal) between the end and middle of the cell- (sub terminal) |
Spores may be differentiated based on shape. What are some of the examples of those shapes. |
Spherical and elliptical( oval) |
What is the procedure for endospore staining |
1. Begin with a heat fixed smear 2. Cover the smear with a paper towel almost as big as slide. 3. Apply malachite green stain 4.Then Steam for 5 minutes 5. Decolorize with distilled water 6.Then counterstain with safranin and |
