Cloning
Dideoxynucleotides halt DNA polymerization at each base, generating sequences of various lenghts that encompass the entire original sequence. Terminated fragments are elctrophoresed and the original sequence can be deduced.
Sanger DNA sequencing
1. random insertion of gene into mouse genome (constitutive)
2. Targeted insertion or deletion of gene through homologous recombination with mouse gene.
Model systems
Used to diagnose chromosomal imbalances (autosomal trisomies, microdeletions, sex chromosome disorders) Can be performed on blood, bone marrow, amniotic fluid or placental tissue.
