Levothyroxine (T4) & Triiodothyronine
Thyroid preparations that activate nuclear thyroid receptors and stimulate gene expression and protein synthesis.

Used for hypothyroidism and myxedema.

Toxicity: tachycardia, heat intolerance, tremors, arrhythmias.
Propylthiouracil
Inhibits peroxidase, inhibits organification of iodide and coupling of thyroid hormone synthesis and also inhibits 5'-deiodinase which decreases peripheral conversion of T4 to T3.

Used for hyperthyroidism.

Toxicity: hepatotoxicity, agranulocytosis.
Methimazole
Inhibits peroxidase, inhibits organification of iodide and coupling of thyroid hormone synthesis.

Used for hyperthyroidism.

Toxicity: teratogenic, CI during pregnancy.
