Define the Median Lethal Dose (MLD or LD50)
Dose which kills 50% of an experimental animal population within specified time after administration.
Define the Therapeutic Index
Ratio between LD50 and the ED50.
= LD50/ED50
*larger the ratio the GREATER the margin of safety

toxic effects which develop only after repeated exposure to a drug for relatively long periods of time
How and why do allergic rxns to drugs occur?
patient has been previosuly exposed AND the drug has functioned as an antigen; genetically-determined predisposition
How do allergic reactions differ from toxic reactions?
allergic rxns are:
-unrelated to dose
-occur only in susceptible person
-not related to pharmacological classification of the drug
What are Iatrogenic disorders?
physician induced disorders from lack of knowledge and understanding of a drug;
e.g. corticosteroids > bone degeneration
e.g. trifluoperazine > tardive dyskinesia
