Baclofen (Lioresal)

MECH: inhibits spinal cord reflexes (GABA receptor agonist), hyperpolarization is involved INDICaTIONS/aDMINISTraTION: drug of choice for MS, effective orally, can be pumped into subarachnoid space surrounding spinal cord SIDE EFFECTS: CNS depression, drowsiness but less than other central acting drugs TOLERaNCE develops, so does must increase, sudden withdrawal can cause seizures and rebound spasticity

BZD prototype: diazepam (valium)

Enhances GABA induced inhibition of neuronal function, inhibits spinal cord reflexes (hyperpolarization involved), effective for spasms and spasticity, high dose required to sedate

Metaxalone (skelaxin)

Used to treat spasms, antianxiety actions, addictive properties, muscle relaxation is linked to general CNS depression

Tizanidine (Zanaflex)

Alpha2 receptor agonist, inhibits transmitter release from spinal cord neurons to decrease motor neuron activity

Dantrolene (dantrium)

MECH: inhibits calcium release from sarcoplasmic reticulum (decrease calcium, inhibits contractions) SIDE EFFECTS: hepatotoxicity and muscle weakness INDICaTIONS: spasms, treatment of hyperthermia, oral or iv

Succinylcholine (alectine)

Stimulated then inhibits skeletal muscle contractions by making it unresponsive to AP, short duration because it is metabolized fast in the liver unless patients have a defect in the plasma esterase, Depolarizing blocker
