Selective medium

Enables transformants to be distinguished from non transformants

Insertional inactivation

Recombinants can be distinguished from self ligated vector molecules because the characteristic coded by the inactivated marker gene is no longer displayed by the host cells

Colony colour

Blue coloured colonies indicate the presence of an active enzyme ie. absence of insert where as colourless colonies indicate presence of an insert (i.e. insertional inactivation)

Recombinant YACs

Are screened by a red-white selection process

Red White Selection

The SUP4 encodes a tRNA that suppresses the Ade2-1 UAA mutation.

In the absence of SUP4 expression the red pigment reappears because the Ade2-1 mutation is no longer suppressed. In contrast the nonrecombinant YAC vectors retain the active SUP4 suppressor. This, red colonies contain recombinant YAC vector DNA whereas the white colonies contain nonrecombinant YAC vector DNA
