Isoniazid Mech and SPec
Mech: Inhibits mycolic acid synthesis
Spec: M. Tuber, M.kansasii, can be used for prophylaxis
Isonizaid pharmacokinetics and SE
Enters CNS, is acetylated, adjust for renal function.
SE: Hepatotox which increases with age. Peripheral neuritis if not given with pyridoxine (vit b6).
Fast and slow acetylators
Fast = japs and escimos
SLow = whites, jews, africans, and scandinavians.

Mech: DNA dependent RNA pol inhibitor.
Spec: M tuber, M marinum, M kansasii, M leprae, MAC.
Rifampin/Rifabutin pharmacokinetics and SE
Enter CNS. Both metabolized by liver, excreted billiary but rifabutin also renal excretion. Have to adjust does if liver failure for rifampin only. Both effect p450, but rifampin is worse.
SE: Hepatitis if given with isoniazid, orange/red urine saliva tears.

Mech: Inhibits mycolic acid
Spec: M. tuber, M, Kansasii, MAC, M. marinum.

No CNS, renal excretion.
SE: Optical neuritis (cant tell green from red), gout
