Reagents: 1 (malachite green/water) counter (safranin O/ethanol/ water)
Acid-fast stain
Dye is forced into cell using heat, remains inside during decolorizing. Acid bacilli will be pink, others blue.
Capsule Stain
Ink forms dark background and capsule stands out as transparent zone.
Fluorochrome Acid-Fast Stain
Decolorized for 2-3 minutes~~counterstained w/potassium permanganate 4-5 minutes.
Metachromatic granule stain
Positively charged methylene ion interacts w/negatively charged phosphate groups. Uses Loeffler's methylene blue, positive cells have blue granules
Spore Stain
Uses 72 hour culture.
Green in pink cytoplasmic background.
