Ink forms dark background and capsule stands out as transparent zone.
Fluorochrome Acid-Fast Stain
Decolorized for 2-3 minutes~~counterstained w/potassium permanganate 4-5 minutes.
Metachromatic granule stain
Positively charged methylene ion interacts w/negatively charged phosphate groups. Uses Loeffler's methylene blue, positive cells have blue granules
Spore Stain
Uses 72 hour culture.
Green in pink cytoplasmic background.
