Aspirin
irreversibly inhibits COX and decreases synthesis of thromboxane and prostraglandins
Use: (low dose) decrease aggregation. (intermediate dose) antipyretic and analgesic. (high dose) anti-inflammatory

Toxicity: gastric upset, chronic use can cause renal failure, interstitial nephritis and upper GI bleed, and Reye's syndrome in kids with viral infections
NSAIDs
reversibly inhibits COX, blocks prostaglandin synthesis
Use: anti-pyretic, analgesic, anti-inflammatory

Toxicity: renal damage, fluid retention aplastic anemia, GI distress, ulcers
celecoxib
reversibly inhibits COX2 sepecifically, which is found in inflammatory cells and vascular endothelium and mediates inflammation and pain

Toxicity: increased risk of thrombosis, sulfa allergy
Acetaminophen
reversibly inhibits COX, mostly in CNS, inactivated peripherally
Use: antipyretic, analgesix, lack anti-inflammatory properties

Toxicity: overdose produces hepatic necrosis; acetaminophen metabolite depletes glutathione and forms toxi tissue adducts in liver
bisphosphonate (-dronate)
inhibits osteoclastic activity and reduces bones formation and resorption of hydroxyapatite
Use: malignancy associated hypercalcemia, Paget's, postmenopausal osteoporosis

Toxicity: corrosive esophagitis (not zoledronate), nausea, diarrhea, osteonecrosis of the jaw
Colchicine
binds and stabilizes tubulin to inhibit polymerization, impairing leukocyte chemotaxis and degranulation,
Use: acute Gout

Toxicity: GI side effects, especially with oral
Probenecid
inhibits reabsorption of uric acid in PCT
chronic gout

Toxicity: decreases penicillin secretion
Allopurinol
inhibits xanthine oxidase, decreases conversion of xanthine to uric acid
Use: chronic gout, lymphoma and leukemia to prevent tumor-lysis associated urate nephropathy
