What causes antimicrobial resistance?
mutation or recombination
What are 4 specific things that we said about antimicrobials?
*can be natural products (antibiotics) or can be synthetic
*act on unique microbial target
*either bacteriostatic or bactericidal
*narrow or broad
Mechanism of microbial resistance
-enzyme degredation
-modification of binding sites
-efflux (pump put of the cell)
-overproduction of target enzyme
-alternation in permeability
note: pleiotropic (multiple mechanisms working together)
what strands of microbes have such complex genomes that they have evolved to a point where they are resistant to all available antimicrobials?
Pseudomonas aeruginosa, Klebsiella pneumoniae and Acinetobacter sp. strains
antimicrobial agents can be either synthetic or natural (antibiotics)...What do synthetic agents consist of?
target selection with use of computer generated molecular modeling
Regarding antibiotics,synergistic combinations are...
when certain combinitions fo antimicrobials have much greater activity than would be predicted by the individual activities of each agent
antagonistic combinations
the combination of antibiotics is significantly less active than either one of the agents alone
Pleiotropic resistance:
multiple drug resistance mechanisms working together

ex: P. aeruginosa strains which have altered porin proteins, beta-lactamase activity and perhaps efflux pumps. If an isolate expresses only one of these resistant mechanisms, the organism will be susceptible. If two or three of these mechanisms are expressed, the organism is resistant.
