Mutations involved

Nod2 mutation is susceptible for chrons. Deficient antimicrobial products in paneth cells. Could lead to accumulation of ileal bacteria and consequently inflammation. Autophagy defects also associated w chrons, due to defects in bacterial killing.


Dysbiosis

Bacteria become pathobionts and can trigger inflammation. This can be caused by impaired t bet signalling in innate immune cells. Bacteria accumulate in t bet negative mice.


Diet can induce dysbiosis. Diet rich in sat milk dat promotes th1 proinflamm response.

Antibodies for ibd

Vedoizumab targets integrins such as a4b7 which is responsible for homing to gut.



Il23 induces th17 cytokine production by ilcs. Th2 response is promoted by il12. Antibody targets p40 subunit of il12 or il23. Antibody targeting il17 didn't work.



Anti tnf first biologic. Prevents proinflamm responses.
