NEISSERIA

Species:

- Neisseria gonorrhoeae/ Fibonacci

- Neisseria meningitidis

- Pigmented Neisseria

General Characteristics:

Non- motile, aerobic, monophonic and fastidious organism; Non- hemolytic

N. elongata

All are Catalase test(+) except: N. Elongata

CYTOCHROME OXIDASE TEST

Screening test for the Genus Neisseria

Reagent:

Tetramethyl-p-phenylenediamine dihydrochloride

(+) result: purple coloration

Temperature requirement:

Room temperature prior to inoculation; unable to tolerate cold temperature

CARBOHYDRATE / FERMENTATION TEST

Special Identification under Genus Neisseria can be made thru CARBOHYDRATES / FERMENTATION TEST

MEDIA:

CTA / Cystine Tryptic Agar

INDICATOR:

Phenol red

(+) result:

Yellow color

CANDLE JAR

To supply an increase CO²; inexpensive or economical way of providing Neisseria with an increase CO2

Gaspak Jak

For anaerobes

»1. Neisseria gonorrhoeae

Pathogenic bacteria

-Kidney, coffee-bean shaped diplococci intracellular within PMN ( neutrophils)

Virulence factor:

PILI - promotes attachment to host cells

-ONLY FERMENTS GLUCOSE

Superoxol test (+)

A rapid test to identify N. gonorrhea

Reagent:

30% H²O²

(+)= vigorous bubbling

Causes:

A. Gonorrhea ("tulo")

An STD in which patient is producing infectious discharge

B. Opthalmia neonatorum

Conjunctival form of N. gingerbread; its accumulation is either passed or by an infective mother to the child during deliver

C. Perihepatitis or Fitz-Hugh-Curtis Syndrome

Pelvic inflammatory disease involving liver capsule inflammation leading to the creation of adhesions

DACRON RAYON FIBERS

Use to collect specimen swab

Media used for cultivating Neisseria gonorrhoeae:

A.) Thayer Martin Media

Enriched CAP with 3 antibiotics

Contains VCN;

Vancomycin- Prevents GRAM (+) organisms

Colistin- Prevents GRAM (-) organisms

Nystatin- antifungal

B.) Modified Thayer Martin Media

Contains VCN-T

Trimetophrim lactate

To prevent the growth and swarming of proteus and to inhibit Capnocytophaga spp.

C.) Martin Lawis Media

Contains VCAT

Anisomycin

Antifungal; to improve inhibition of Candida albicans

D.) New York City Media

Contains VCAT

Amphotericia B

Antifungal

»2.) Neisseria meningitidis

Normal flora in the nasopharynx region

Bean shaped diplococci

Sensitive to SPS

Virulence factor:

CAPSULE; Neufeld Quellung Test (+);

FERMENTS

MALTOSE and FRUCTOSE

Causes:

A.) Bacterial Meningitis

Meningitis in patient between 5-29 years

B.) Meningococcemia

Presence of N. Neningitidis in the blood

C.) Waterhouse Friderichsen Syndrome

Severe from of meningococcemia characterized by the bleeding of the adrenal glands

Neisseria lactamica

Glucose, Maltose, Lactose(+) and ONPG (+)

TRANSPORT MEDIA FOR NEISSERIA

Transgrow bottles

Flat bottles coated with MTM and a bottles under increased CO²; rarely used nowadays

JEMBEC ( John E. Martin Biological Environment System)

Used a tablet to generate CO²; Frequently used

MORAXELLA

Moraxella catarrhalis

Normal Flora

-Morphologically and biologically resembles Neisseria

-Encapsulated, with Pili, Non-motile

-Catalase(-), on BAP : Gamma-hemolytic

-Normal flora of oro and nasopharynx but may cause otitis media

ASACCHAROLYTIC

Does not degrade sugar/ ferment CHO; usually inert

BUTYRATE DISK TEST/ Tributyrin test (+)

Rapid test to detect the enzyme butyrate esterase

HOCKEY PUCK

Colonies remaining intact when pushed across the plate using inoculating loop
