Mimivirus "Mimicking microbe"

First giant virus discovered. Found in Amoebae. Isolated from A. polyphaga. Icosohedral capsid. Enveloped. 400 nm (comparable to small bacteria). Linear dsDNA genome.

Mimivirus replication

Replicates in cytoplasm. Orchestrated by its own genes.

Mimivirus phylogeny

based on 8 conserved viral proteins.

Megavirus

Related to Mimivirus. 77% of genes have homolog to Mimivirus. 6.5% larger genome than Mimivirus.

Divergence between Mega and Mimivirus

Primarily due to accumulation of point mutations.

Genes unique to one or the other usually found

clustered near the ends of the chromosome

The genomes also differ in

expansion or reduction of paralogous gene families.

Stargate structure

Found in both Mimi and Megavirus. At the vertex of the capsid. Structure opens allowing the release of viral DNA into the host cytoplasm.

1st hypothesis of Mimiviridae origin.

Large genomes are a result of extensive transfer from host, bacteria, and other viruses

2nd hypothesis of Mimiviridae origin.

Most genes originated from and even more complex viral ancestor, itself derived from and ancestral cellular genome.
