How do to you set up a photosynthometer?

-rate proportional to volume of oxygen produced in a known period of time.

-Cut a piece of an aquatic plant e.g. Elodea and to make sure bubbles emerge from cut stem place upside down in test tube containing NaHCO3.

-gas given off collects in capillary tube and syringe is used to put air bubble on the scale.

-Length x Pir^2

Limitations and improvements of photosynthometer

-There may be dissolved nitrogen or CO2 from photorespiration in the gas collected.

-Temperature may fluctuate thus use thermostatically controlled water bath and thermometer.

-Other sources of light, use a low energy bulb or darkened room

-Make sure airtight so no bubbles in the collecting tube.

-Improve experiment by adding a coloured filter to lamp.

How to set up leaf disc experiment and improvements?

-Drinking straw to cut out discs from cotyledons (seed leaf). Pick 5/6 and place in syringe with NaHCO3. Hold syringe upright and pull the air out which pulls air out of air spaces in spongy mesophyll and density increases (sink to bottom). Disks added to beaker solution with Light source. Time reaction to get to top (decrease in density) due to release of O^2.

-Improve: same size (surface area),p same area of plant (no. Of chloroplasts), temp (thermostatically controlled water bath), excess CO^2.
