SX of Parkinson's Disease caused by...
Deterioration of the dopaminergic neurons in the substantia nigra pars compacta, resulting in depletion of dopamine along the nigrostriatal pathway to neurons in autonomic ganglia, basal ganglia & spinal cord causing neuro deficits.
SX of PD are...
- muscle tremors,
- slowness of movement in performing daily activities (bradykinesia),
- muscle weakness with rigidity, and
- alterations in posture and equilibrium
What neurotransmitter pathway is affected in PD?
Dopaminergic pathway (inhibitory neurons) in the substanstia nigra and corpus striatum (neurotransmitter ratio shifts toward decreased DA and increased Ach)
- Increased ACh causes PD sx
Drug therapy for PD
1. Rasagiline or selegiline to slow deterioration of dopaminergic nerve cells
2. Carbidopa-devodopa, ropinirole, pramipexole amantadine or entacapone to enhance dopaminergic activity
3. Anticholinergic agents to inhibit excess cholinergic activity
