What activates platelets?

Bernard-Soulier syndrome- GP1b deficiency disrupts platelet adhesion to collagen
Glanzmann's thrombasthenia- deficiency in GPIIb disrupts platelet aggregation |

What does vitamin K do?
|
gamma-carboxylation of pro-coagulation factors: "1972CS" --> 10, 9, 7, 2, protein C and protein S
(carboxylation is necessary for calcium binding) |

What does warfarin do?
|
blocks gamma-carboxylation of glutamate residues in the 1972 factors. Found in rat poison.

How does thrombin regulate the coagulation cascade?
|
Thrombin activates factors 5, 8, and 11 while it binds to thrombomodulin (which then causes protein C/S activation and factor 5 & 8 degradation leading to coagulation inhibition)

What does ATIII do?
|
Anti-thrombin III irreversibly inactivates thrombin and is enhanced by heparin
|

|
A= factor 8 deficiency, X-linked, joint and muscle hemorrhage + bruising and prolonged bleeding
B= factor 9 deficiency, 1/10, prolonged coagulation time |

Describe factor 13 deficiency
|
Factor 13 is activated by thrombin to become active plasma transglutaminase. . Def result in neonatal umbilical cord bleeding, intracranial hemorrhage, soft tissue hematomas
