What is the MOA of doxorubicin?
trade names: Adriamycin, Rubex

category: anthracycline antibiotic

MOA: complex, DNA intercalater --> inhibits topo II, prevents mRNA synthesis.
What is doxil and what is the most common side effect?
pegylated liposomal doxorubicin

increased concentration in the skin --> hand foot syndrome in 50% of patients
What are the toxicities of doxorubicin?
N/V, arrythmias, neutropenia, alopecia, hand foot syndrome

CHF, dilated cardiomyopahty. Characterized by a dose-dependent decline in mitochondrial oxidative phosphorylation. Reactive oxygen species, generated by the interaction of doxorubicin with iron, can then damage the myocytes, causing myofibrillar loss and cytoplasmic vacuolization.
palmar plantar erythrodysesthesia is also known as what?
hand-foot syndrome
What is MOA of epirubicin?
anthracycline, DNA intercalater. prevents DNA and RNA synthesis. Triggers DNA cleavage by topo II. thought to bind to cell membranes and plasma proteins.

faster elimination and reduced toxicity compared to doxorubicin.
Trade name of epirubicin?
Marketed under Pfizer as Ellence
What is MOA of cyclophosphamide?
nitrogen mustard alkylating agent.

prodrug that is converted into active form by the liver.
Trade names of cyclophosphamide?
Cytoxan, Endoxan, Neosar, Procytox, Revimmune
What is the MOA of paclitaxel?
microtubule stabilizer --> interferes with the normal breakdown of microtubules. inhibits mitosis.
What is tradename of paclitaxel?
Taxol
Taxanes refer to what?
paclitaxel and docetaxel (and others)
What is the tradename of docetaxel?
Taxotere
What are the toxicities of taxanes?
N/V, loss of appetite, change in taste, thinned or brittle hair, pain in the joints of the arms or legs lasting 2–3 days, changes in the color of the nails, tingling in the hands or toes.

More serious side effects such as unusual bruising or bleeding, pain/redness/swelling at the injection site, change in normal bowel habits for more than 2 days, fever, chills, cough, sore throat, difficulty swallowing, dizziness, shortness of breath, severe exhaustion, skin rash, facial flushing and chest pain can also occur. A number of these side effects are associated with the excipient used, Cremophor EL, a polyoxyethylated castor oil. Allergies to drugs such as cyclosporine, teniposide and drugs containing polyoxyethylated castor oil may indicate increased risk of adverse reactions to paclitaxel. Dexamethasone is given prior to beginning paclitaxel treatment to mitigate some of the side effects.
What is MOA of lapatinib?
SMTKI of Her2
What is MOA of trastuzumab?
MAb against extracellular Her2 receptor.
What are the MOAs of carboplatin, cisplatin and oxaliplatin
DNA alkylating agents
What is the MOA of gemcitibine?
nucleoside analog. replaces cytidine during DNA replication.
