Valacyclovir is valine analogue of acyclovir
-Allows for 3 to 5 times greater bioavailability than oral acyclovir
Purine nucleoside analogs
-Suppress synthesis of viral DNA
Blocks polymerase enzyme that builds DNA and is incorporated into viral DNA
– Resistance-more common in allograft and HIV

MOA not fully understood, stops entrance of virus and affects replication.

Adverse effects
CNS-seizure, depression, hallucination
Drugs for Influenza:
Oseltamivir [Tamiflu]
Inhibits neuraminidase, an enzyme known to cleave the budding viral progeny from its cellular envelope attachment point (neuraminic acid) just prior to release
Current drug of choice for avian and swine flu
Must be use early in disease for maximum benefit

ADR:
Gastrointestinal: Vomiting (2% to 15%)
Gastrointestinal: Nausea (3% to 10%), abdominal pain (2% to 5%)
