What are the two ways to prepare slides? |
Dry mount and wet mount. |
What is a wet mount? |
Put a drop of water onto the slide. Add a thin slice of the specimen and add the dye. Artefact = bubble |
What is a dry mount? |
Take a thin slice of the specimen and put onto the slide. |
What is a light microscopes resolution? |
0.2 |
What is a TEM microscopes resolution? |
0.0002 |
What is a SEM microscopes resolution? |
x0.002 |
How does a laser scanning confocal microscope work? |
Specimen is fluoresently dyed, a laser is then scanned over it. The light which shines back is concentrated into a pinhole and onto a detector. |
What are the two types of electron microscopes? |
SEM and TEM |
How does a TEM microscope work? |
Transmits a beam of electrons through a specimen. The denser parts absorb more electrons. |
How does a SEM microscope work? |
Scans a beam of electrons across the surface of a specimen. |
How do you go from micrometers into mm? |
Divide by 1000. |
