Methotrexate

Type: folic acid analog


Use: breast cancer, osteogenic sarcoma, ALL, Burkkits lymphoma, bladder cancer


Adverse effects: nephrotoxicity, mucositis, myelosuppression, pulmonary and hepatotoxicity

5-fluorouracil/floxuridine

Type: pyrimidine analog


MoA: complexes with folic acid and inhibits thymidylate synthase


Use: breast cancer, colorectal cancer, hepatoma


Adverse effects: toxicity when used with folinic acid/leucovorin in people with dihydropyrimidine dehydrogenase deficiency

Cytaribine

Type: pyrimidine analog


Use: AML


Adverse effects: neurotoxicity, myelosuppresion, megaloblastic anemia

Cyclophosphamide

Type: CCNS alkylating agent


PK: must be activated by liver


Use: solid tumors, CHOP regimen, neuroblastoma, SLE


Adverse effects: nephrotoxicity, chemical cystitis, SIADH

Bulsufan

Type: CCNS alkylating agent


Use: CML & AML


Adverse effects: pulmonary fibrosis, hyperpigmentation, adrenal suppression, cataracts, thrombocytopenia

MoA: cross links DNA-> breaks

MoA: causes breaks in DNA by inhibiting topoisomerase II and by free radical formation

Adverse effect: peripheral neuropathy, constipation (paralytic ileus)
