Vincristine
Mitotic Spindle Poison, prevents microtubules from forming--> thus stopping the mitotic spindle. Causes peripheral neuropathy
Paclitaxel
Mitotic Spindle Poison. Prevents Microtubule dissociation freezing the spindle in place. Causes peripheral neuropathy
Cyclophosphamide
Mustard Gas prototype. Binds DNA, prevents transcription and duplication. Need P450 to be activated. Very Immunosupressive
Cisplatin
DNA alkylating agent.Ditrubutes into tissues better than cyclophosphamide.
Doxorubican=adrimycin
Topoisomerase inhibitor. Used in breast cancer. Break dna when topoisomerase tries to unwind it. Cardiac toxicity common.
5-floro-uracil
5FU prevents thymidilate synthetase from adding a methy group to uricil, thus creating thymidilate. All serves to slow transcription.
Methotrexate
Methotrexate inhibits dihidrofolate reductase, inhibiting the formation of THF that donates the methyl group for thymidilate synthatase.
Slows transcription |
Bevacizumab
VEGF Target-er. O2 drops in tumor, VEGF gets released, causes angiogenesis. $70,000, 5 month improvement. Causes Hypertension. Signifigant Use in Macular Degeneration.
Cetuximab
Anti EGFR (=HER1) antibody. Often used for corectal cancer. Works better in wild type Ras population.
Traetuzumab
Anti HER2 Antibody--> Her-2 mutation bad news in breast cancer, but signifies weakness to Drug. Causes cardiac side effects.
Rituximab
Anti CD20 B Cell antigen, Leukemias Probably
Geftinib, Erlotinib
EGFR- Kinase inhibitor. Resistnce caused by a mutation in T790M. Cause itchiness like Cetuximab, same Ras relationship as well.
Lapatinib
Very Specific for Her-2 kinase
Sorafenib, Sutinib
VEGFR-r internal kinase targeter. Thats it.
Imatinib
Abl/Bcr targeter. Great at first but resistance is likely.
Vemurafenib
Targets BRaf which is downstream of Ras. Used in metastatic melanoma. Only really gives you like 6 more weeks
Ipilimunab
Blocks the CTLA4 receptor on T cells so that tumor cells can't use it to suppress growth. Anti PD-1 does a similar thing to PD1. Also used in Metastatic Melanoma. Inflammatory, autoimmune side effects.
Bortezomib
Proteasome ihibitor. Cells cannot degrate protein and become apoptotic. Does cause system wide side effects, good additional drug.
Azathioprine
T cell cycle inhibitor,
Sirolimus
Interferes with IL-2 pathway in T Cell
Cyclosporine/Tacrolimus
Interferes with calcenurin transduciton of the TCR pathway.
Basiliximab, Daclizumab
Anti IL2-R antibody.
Infliximab
Anti TNF antibody
