first person who created non-reusable microscope (1600s)
Anton von Leewonhoek
focuses light rays that pass through the specimen to produce a magnigfied image
(staining) |
compound light microscope
conatains magnifying lens. it has a magnifiacation of 10x
(eyepiece) |
Ocular
it lays between the ocular and arm; allows light to pass to your eyes from objective lenses
(prism back ups) |
body tube
support for body tube. it connects the body tube to the base
arm
where you put a slide to be viewed
stage
allows light to pass through the slide
stage opening/aperture
large knob that moves the stageor body tube upward or downward to help focus the image
coarse adjustment knob
small knob that brings the image into sharp focus
fine adjustment knob
bottom of the microscope; it supports the entire microscope
base
holds the slide in place
stage clips
holds the objective lense
revolving nosepiece
part that regulates the amount of light intensity
diaphragm
rotating disc with several size holes
wheel
has a lever with greater selection of holes
iris
has slots to recieve filters that can remove specific wavelengths of light
condenser
provides largest field of view and lowest magnification (4x)
scanning power
higher magnification (10x)
low/medium power
highest magnification(40x) smallest field of view
oil(100x) |
high power and oil emersion
what you see when you look in the occular; low magnification has a larger view than high magnification
field of view
is calculated by multiplying magnification of the eyepiece and objective magnification
total magnification
where you get detail, if you go past it the image will be blurry
limit of resolution
how to clean a microscope
wipe the lense with lens paper. sont reuse a piece of the lense paper
how to carry a microscope
one hand on the arm and one on the base; hold it upright
how to store a microscope
put it on the lowest possible onjective position; lowest stage; dust cover on; cord wrapped around the base
how to place a slide on a microscope
put the slide centered on the stage and clip it down
what are the 8 steps to focus a microscope
1. make sure the lenses are clean
2. put the side on and clip 3. put the scanning objective in place 4. focus the course adjustment knob 5. focus the fine adjustment knob 6. switch to low power and readjust 7. repeat with medium 8. when finished reverse the steps |
how to prepare a wet mount (5 steps)
1. clean slide
2. place a thin layer of the specimin in the center of the slide 3. use pipette to put a tiny drop of water in teh middle of slide 4. lower cover slip on to slide 5. remove exess water with paper towel |
