freeze fracture
technique in which tissue is rapidly frozen in liquid nitrogen then fractured for further treatment before microscopic examination.

shadowing
technique in which fractured pieces of specimen within a vacuole chamber are exposed to , and partly covered by heavy metal that is evaporated; the metal film reflects the contours of the parts that are covered and the parts are then covered with a layer of carbon atoms that is transparent to electrons. this speciman fragments are dissolved away leaving metal replicas that can be examined with an electron microscope.
