AHG
antiglobulin test
-detects IgG antibodies and complement proteins that have attached to the rbcs but have not resulted in agglutination
AHG principle
-animals injected with human antibodies, and produce anti-human globulins, or anti-c3b/c3d
-we harvest these anti human globulins and use these as reagents
-the ahg will bind to the Fc portion of sensitising antibodies on rbcs
-these form bridges resulting in agglutination
Coombs control cells
-commercially prepared rbc with igG attatched
-added to all negative ahg tests
-reactions 2+ or stronger
Polyspecific reagents
-contains antibodies to human IgG and C3
-primarily used in DAT
Monospecific Reagents
-has either Anti-IgG or Anti-C3 only
DAT
Direct Antiglobulin Test
-detects IN VIVO bound antibody
-Hemolytic Disease of Newborn, transfusion
-Autoimmune Hemolytic Anemia
-Drug induced hemolytic anemia
IAT
Indirect Antiglobulin Test
-detects IN VITRO bound antibody
-need serum
