ACTH 1-39 is a synthetic polypeptide hormone that consists of 39 amino acids. ACTH 1-39 is commonly referred to as Adrenocorticotropic hormone, corticotrophin, or ACTH. The molecular formula for ACTH 1-39 is C207H308N56O58S1 and a molecular mass of 4541.11 kilo Daltons. The structure of ACTH 1-39 corresponds to the full-length structure of ACTH. In this study, the researchers sought to determine the role played by the fetal hypothalamo-pituitary axis in regulating adrenocortical growth and steroidogenesis within sheep fetus that is in late gestation period. The researchers observed a significant rise in the levels of ACTH 1-39 in the plasma between 120-125 and 136-142 days, but did not observe any change after the fetal hypothalamus and pituitary were surgically disconnected at 106-120 days of the gestation period. The results did not show the effect of both HPD and gestation age on the circulating concentrations of pro-ACTH and ACTH-containing the precursors pro-ACTH and pro-opiomelanocortin. Analysis of the fetal sheep adrenal revealed an increase in the relative abundance of mRNAs from steroidogenic enzymes CYP21A1 and CYPIIA1 during the period between 130-135 and 136-140 days of the gestation and was sustained at high levels after 141 days gestation. On the other hand, a significant increase in relative abundance of CYP17 mRNA was noted after 141 days gestation. No changes were noted in the relative abundance of the adrenal 3 beta-HSD mRNA between 130-145 days. The researchers harvested 44 chambers of perifused pituitaries from mature ewes and took them through two challenges with GnRH . They then expressed their results as percentages of the baseline values obtained from each chamber. In their observations, both ACTH 1-39 and CRH did not affect tonic LH secretion compared to the controls or the first GnRH-stimulated LH secretion. In the second challenge, the response to LH to the second GnRH challenge was inhibited significantly. the two doses of ACTH 1-39 showed significant inhibition to the degree of GnRH self-priming, similarly to CRH when the researchers expressed the second LH response as a percentage of the initial LH response. The researchers concluded that both CRH and ACTH 1-39 had no effects on the secretion of LH induced by tonic, but they inhibited the secretion of LH that had been induced by GnRH by avoiding GnRH self priming.
