Objectives: In the present study we made an attempt to evaluate the association of Plasminogen Activator Inhibitor-1(PAI-1)4G/5G polymorphism with oxidative stress markers Malondialdehyde (MDA) and High sensitivity C-reactive protein (hs-CRP), fibrinolysis marker PAI-1and lipid profiles. Subject and Methods: Blood was drawn and DNA extracted from 90 subjects (46 cases and 44 controls). The 4G/5G polymorphism of PAI-1 was amplified using specific primers. Amplified products were visualized by staining with Ethidium bromide after electrophoresis in agarose. Risk factors PAI-1, MDA, hs-CRP and lipid profile were compared between 46 angiographically diagnosed coronary artery disease(CAD) patients and 44 age/sex matched healthy subjects. It has also reported that high level of PAI-1 is associated with the impairment of vascular wall and endothelial dysfunction. (4).Human PAI-1 gene is located at chromosome 7q22, and various genetic polymorphism have been identified in its promoter region (4). A common insertion/deletion of guanine have been seen in this region (3). The deletion of the allele (4G) is associated with increased PAI-1 plasma level due to inability of transcriptional repressor protein for binding to this polymorphic sequence (5).
C - reactive protein (CRP) is a member of Pentraxin family, which plays a key role in the innate immune response. CRP is the most sensitive acute reactants protein and its concentration increases rapidly during inflammatory process (6-8). This protein has been shown to have prognostic value in patients with acute coronary syndromes and play different roles in pathogenesis of atherosclerosis (7).
MDAa marker which is commonly used to evaluate the lipid peroxidation. Its assessment is considered as an undependable marker of oxidative damage, therefore MDA could be a useful indicator for the identification of patients with CAD (7).
