The handkerchief symbolizes the ocular proof Othello needs to know that Desdemona is cheating on him. Iago being his manipulative self is trying to make Othello focus on the handkerchief rather than what Othello wants to see, them kiss, or in bed together. So Iago keeps dropping hints about the handkerchief. Once Othello sees that it’s the handkerchief that he gave to Othello he says. This shows that Othello is falling into Iago’s trap and believes what he’s saying. Later in the scene, Iago plans that Othello will overhear Cassio talking about Bianca, his prostitute girlfriend, hoping Othello will think he is talking about Desdemona. Iago gets lucky, during this Bianca shows up and she has the handkerchief with her. She says "There; give it your hobby-horse: wheresoever you had it, I 'll take out no work on 't" (4.1.154-155). She has decided that she was a fool to agree to copy the handkerchief, and a fool to accept Cassio 's story that he found it in his room. She now throws it back at him, tells him that he should give it to the whore he got it from, and declares that no matter where he got it, she 's not about to copy it. As soon as Cassio is gone, Othello comes out from hiding, asking how he should kill Cassio.
