3.1 The isolation of Aeromonas hydrophila When using selective medium developed by Rimler and Shoots (1973), we obtained 95 bright yellow colour isolates with white edge which lead to the alleged as A. hydrophila. Of 95 isolates were further screened using SNI 7303 (2009) methods which produce 56 isolates. The isolates were further screened using Dorsch et al. (1994) and Cascón et al. (1996) method. 3.2 Bacterial DNA isolation and Identification. DNA amplification using Dorsch method…
The oldest forms of life are single-celled organisms known as microbes. There are many different types of microbes that are found all around the world, and they play an important role in the environment, in the body of animals, and even in the health of plants. The specific type of microbe investigated in this paper is the gram positive bacterium known as Kocuria rhizophila, which has a bright yellow color that simplifies identification. The main objectives of the paper are the following:…
the experiment. This included, a microscope, three glass microslides, alcohol, caffeine, and water, four pippets, then finally three water fleas. After our workspace was ready we took the first flea and placed the flea on a glass plate then carefully latched the plate onto the microscope. Once the third team mate located the water flea on the microscope, I placed one single drop of water from…
To change solid agar into liquid, get 4 Petri dishes out and ready for use. Next, agar needs to be a boil, either on heating pot or stove, not in a microwave. Agar bottle needs to be placed inside the pot, with water in it to be a boil. Water level needs to be right under the label on the agar bottle, with the cover losing, or it will explode. Agar needs to be boil for about 30 to 45 minutes, until agar liquefied. Then pour agar inside the petri dish, about half full. Make sure agar is fully…
ABSTRACT: This study was carried out to evaluate the effect of pretreatments prior different drying methods and temperatures on compositional quality of the dried mushroom. Oyster mushroom (Pleurotus ostreatus) was cultivated on rice substrate and collected samples were pretreated (steeped or blanched) by different solutions (0.1% sodium metabisulfite and 0.1 and 0.5% citric acid) and dried using oven at 50°C and 60°C, sun-drying at 30-35°C and microwave drying at 2.45 GHz until reached to…
The Effect of Carbon Sources on Yeast Growth Introduction Biofilms are encountered every day through a multitude of ways whether it’s by dental plaque on our teeth, in our showers or on medical equipment. (Hall-Stoodley 2004). A biofilm is formed when single bacterial cells stick to each other (Schussler 2014). Biofilms as a group converse and work together as a complex unit which allow them to be effective in growing, surviving and reproducing (Liu et al. 2015). Moreover, yeast biofilms are an…
Agar blocks of the stem TR-S1 endophyte were cut from the pure strain actively growing on PDA Petri dishes and transferred to 1 L Erlenmeyer flasks containing a 200 g sized recipe of Corn Grit Agar(CGA). TR-S1 endophyte was incubated at room temperature at 26-28C for 21 days. Fungal metabolites were then extracted by acetone which was then partitioned with ethyl acetate and methanol respectively. TLC analysis of the ethyl acetate fraction revealed the characteristic blue spots of polyketides…
To determine the effect of the proximity of the dam on fish distributions downstream, I will collect data at three locations along the Monongahela River, moving downstream from the dam (Site 1, Site 2, and Site 3 in Figure 1.A). I will repeat this two times on two different days of similar weather patterns for a total of $110 in anticipated costs. At each site, I will collect a variety of variables. For starters, I will collect an indication of water clarity using a secchi disk at each site for…
The purpose of this experiment was to swab a chosen object, isolate a microorganism, and identify the microorganism through a series of morphological, physical, and biochemical tests. One chose to swab David Phelps’ pastor dreamer, which is one of the larger statues on The University of Oklahoma’s campus. The nose of this statue was swabbed and is where one’s environmental isolate originated. Through morphological, physiological, and biochemical experiments, one was able to gather results that…
We first evaluated whether Honokiol treatment was toxic to the APP-CHO cell line. Cells were treated with various concentrations of Honokiol (0.5 μM, 1 μM, 2 μM, 5 μM, 10 μM and 20 μM) for 24 h and cell survival was determined by Prestoblue® assay (Fig.1A). As shown in Fig.1A, when exposed to Honokiol concentrations of 10 μM or lower, the viability of APP-CHO cells was the same as untreated control cells. However, a significant decrease in cell viability was observed with 20 μM Honokiol…
