Need: Nigrosin and two slides 1. add 1 drop nigrosin on left 2. inoculate 3. Place 2nd slide in front of nigrosin, move over it, and move it back 4. air dry |

Gram Stain |
Need: Crystal Violet, Gram's Iodine, Ethanol, Safranin 1. One drop of distilled water (small) in middle of slide 2. Innoculate with bacteria, air dry, then heat fix 3. Crystal Violet- 1 min 4. Rinse with water 5. Gram's Iodine- 1 min 6. Decolorize with ethanol~ 10-15 secs 7. Safranin- 1 min 8. Rinse with water 9. Blot with bibulous |

Capsule Stain |
Need: Congo Red and Manevals 1. Add drop of Congo Red on left side; innoculate; use slide to spread like with negative 2. Air dry 3. Manevals- 1 min 4. Rinse with water 5. Blot |

Acid Fast |
Need: Carbolfuchsin, Methylene Blue, Acid Alcohol 1. Drop of water in middle; innoculate; air dry; heat fix 2. Cover with bibulous paper (small strip so just covers slide) and place on hot plate 3. Cover with carbolfuschsin (8-10 mins) (don't let dry) (put paper in biohazard when done) 4. Rinse with water then acid-alcohol (til runs clear) then water again 5. Methylene blue- 1 min 6. Rinse with water and blot |

Endospore |
Need: Malachite Green and Safranin 1. Drop of water in middle; innoculate; air dry; heat fix 2. Bibulous paper on hot plate 3. Soak with Malachite Green (don't let dry) 8-10 minutes; dispose of bibulous in biohazard 4. Rinse with water 5. Safarin- 1 min 6. Rinse with water and blot |
