Drugs often associate with proteins. Unbound drug can move between compartments until equilibrium is reached however this is dependant on permeability, ph, binding capacity etc.

Metabolism

1st pass metabolism - 1st pass through liver, sometimes very dramatic so certain drugs much better IV

Cytochrome P450

Group of enzymes that metabolise drugs in the liver by several processes including oxidation and hydroxylation.




Some drugs are pro drugs and only become active once metabolised e.g aspirin.

Excretion

Most drugs secreted by kidneys. Small number excreted in the bile.

Dronotropic

When a drug increases the speed of the conduction pathway.
