Pharmacokinetics and dynamics |
Pharmacokinetics = what the body does to drugs Pharmacodynamics = what does the drug do to the body. |
ADME |
Absorption Distribution Metabolism Excretion |
Half life |
The time it takes to remove half the current conc of a particular drug |
Distribution of drugs |
4 main compartments Blood plasma Extra cellular fluid Intracellular fluid Other - CSF, foetus, synovial fluid etc Drugs often associate with proteins. Unbound drug can move between compartments until equilibrium is reached however this is dependant on permeability, ph, binding capacity etc. |
Metabolism |
1st pass metabolism - 1st pass through liver, sometimes very dramatic so certain drugs much better IV |
Cytochrome P450 |
Group of enzymes that metabolise drugs in the liver by several processes including oxidation and hydroxylation. Some drugs are pro drugs and only become active once metabolised e.g aspirin. |
Excretion |
Most drugs secreted by kidneys. Small number excreted in the bile. |
Dronotropic |
When a drug increases the speed of the conduction pathway. |
