The GasPak system

Is useful for culturing anaerobic bacteria on standard microbiological medium because the gas pack generates carbon dioxide and hydrogen

The hydrogen will combine with the oxygen present in an anaerobic jar to produce...

water

The gaspak system is the best method for determining...

..the oxygen requirements of unknown organisms

Candle jar

Useful for culturing organisms which prefer reduced oxygen levels and increased carbon dioxide levels

-NOT an oxygen-free system

This is the best method of culturing a microaerophile

Obligate anaerobe

A microbe that cannot tolerate oxygen and will be killed in his presence.

This type of organism is killed because it lacks the enzymes necessary to eliminate toxic oxygen products.

obligate aerobe

A bacterium that requires atmospheric levels of oxygen for normal growth

Facultative anaerobe

A microorganism that can grow with or without oxygen but usually grows faster (produces more ATP) in its presence

Aerotolerant anaerobe

A microbe which will survive but will not grow in the presence of oxygen.

Microaerophile

An organism which requires reduced levels of oxygen

Sodium thioglycollate broth

Contain substances which chemically combined with oxygen making it unavailable.

Anaerobes will grow in this medium.

Aerobes can grow but only in the upper layers of this medium

In an anaerobic chamber

A gas pack of chemicals is activated using 10 mL of dh2o (distilled water.) The gas pack releases CO2 an H2 gases. Most anaerobes will prefer elevated CO2 levels. There's a mesh capsule in the lid which contains a catalyst called Palladium,in the presence of this, H2 gas is released from the gas pack and any O2 in the chamber chemically reacts forming H2O.
