What is a pure culture?
A culture of only one genus and species of an organism.
What are the techniques to isolate a pure culture called?
Pure culture techniques.
How do you keep organisms from the air, hands, table, etc from contaminating the culture?
By using antiseptic or sterile techniques.
What is an antiseptic or sterile technique?
Procedures to keep unwanted microorganisms from contaminating either sterile materials or pure cultures.
How are culture media and glassware sterilized?
They are sterilized in an autoclave at 121 degrees C, at 15 lbs of pressure for about 15 minutes.
What nutrients must be provided so microorganisms can be grown in a library?
Carbohydrates, lipids, proteins and vitamins. As well as oxygen and the proper humidity and heat.
What is a broth?
A liquid medium that organisms can be grown on.
What is agar?
A hardening agent added to broth to create a solid surface organisms can grow on.
What is a slant?
Liquid agar poured into a test tube and allowed to harden at an angle.
What are three simple methods of isolating a pure culture?
Streak plating with a loop, spread plating on a solid medium with a glass spreader, and pour plates.
What is the purpose of spread plating and streak plating?
To isolate the individual bacterial cells on a nutrient medium so that during subsequent incubation, a cell grows and divides sufficently to create a single colony
How is a spread playing prepared to encourage individual colonies?
A diluted culture must be used so the number of bacteria per unit volume is decreased.
How is streak plating used?
Streak plating isolates individual bacterial cells on the surface on an agar plate using a wire loop.
