What is the purpose of the Urease Test?
To differeniate organisms based on their ability to hydrolyze urea with the enzyme urease.
How is Urea produced?
It's a product of decarboxylation of certain amino acids. It can be hydrolyzed to ammonia and carbon dioxide by bacteria containing the enzyme urease.
Why was the Urea agar formulated?
To differeniate rapid urease + organisms from the slower urease + and urease - bacteria.
What are the 2 differences between urea agar and urea broth?
Urea broth only has one nutrient source, trace of yeast extract. It also contains buffers strong enough to inhibit alkalinzation of the medium but the rapid urease + organisms.
what is the purpose of the Gelatinase test?
to derernmine the ability of a microbe to produce gelatinases.
What are gelatinases?
Protein dervived from collagen, Gelatinases comprise a family of extracellular enzymes produced and secreted by some microorganisms to hydrolyze gelatin.
What does SIM mean?
Sulfur-Indole-Motility
What kind of medium is SIM?
Combination Differential
What is a stab inoculation and why is it used?
When you stab the agar with the loop and it is used to produce anaerobic/motility microbes for transfer to another medium. It's used for differential mediums.
What is Enviromental Microbiology?
The study, utilization, and control of microorganisms living in marine, freshwater, or terrestrial habitats.
What is the purpose of the membrane filter technique?
commonly used in combination with other tests to identify the presence of fecal coliforms.
What is the microbial soil count for?
To isolate and demonstrate the variety if populations residing in soil.
What is meant by food microbiology?
Study, utilization, and control of microorganisms in food such as yogurt, wine, beer, sauerkraut, buttermilk, etc.
What is the purpose of the methylene blue reductase test?
This test is helpful in differentiating the enterococci from other streptococci. It also test for the presence if coliforms in raw milk.
What is the purpose of the Snyder Test?
to measure dental carries susceptibility caused by primarily by lactobacilli and oral streptococci.
What is the color of a + synder test?
yellow
What is antimicrobial testing?
Used to measure the effectiveness if antibiotics and toehr chemotherapeutic agents on pathogenic microorganisms.
What are antibiotics?
natural antimicrobial agents produced by microorganisms
What is the Kirby-Bauer Test used for?
for measuring of the effectiveness of antimicrobics against pathogenic microorganisms.
What is the zone if inhibition?
The sensitivity of the bacteria to the specific antimicrobial agent and the point at which the chemicals MIC is reached.
What is MIC?
Minimum inhibitory concentration
What is a synergistic effect?
A clear area between the disks extending beyond the perimeters of the otherwise circular zones.
What are pyrimidine dimers?
Cytosine-cytosine, cytosine-thymine, thymine-thymine
What is photoreactiviation?
Light repair in which the repair enzyme, DNA photolyase, is activated by light and simply monomerizes the dimer by reversing the original reaction.
