N-Acetyl-L-cysteine (NALC) & NaOH

Most commonly method used.

Trisodium phosphate (TSP) and benzalkonium chloride (Zephiran)

TSP liquefies sputum rapidly but required a long exposure for decontamination.

Z shorten the exposure time and effectively destroys many contaminants with little bactericidal effects to bacilli.

Oxalic acid

Oxalic acid for sputum collected from patients with cystic fibrosis -eliminatr mucoid strains of Pseudomonas aeruginosa.

4% Sodium hydroxide (NaOH)

Strong alkali acts both mucolytic and antibacterial agent.

Exposure time must be limited used with caution because it can kill the Mycobacteria.
