Uses

To identify the microorganism.

Antibiotic susceptibility testing.

Basic constituents

Water

Electrolyte,

Peptone

Meat extract

Blood/serum

Agar

Peptone

- Mixture of partially digested proteins

- Obtained from lean meant, proteins like heart muscle, casein or fibrin, digested with proteolytic enzymes.

- Contains proteoses, amino acids, polypeptides, phosphates, minerals and accessory growth factors.

- Special types - neopeptone and proteose peptone

Accessory growth factors

Nicotinic acid,

Riboflavin

Meat Extract

Available as lab lemco

Contains protein degradation products, inorganic salts, carbohydrates, growth factors.

Blood

Used for enriching the media

5 to 10% defibrinated sheep blood is used.

Agar

Prepared from sea weed/algae - geladium species.

Contains long chain polysaccharide, protein like material and inorganic salts.

Concentration - 2 to 3%

Melts at 98°C

Solidifies at 42°C

Does not provide nutrition but acts as a solidifying agent.

Brain-Heart Infusion

For the culture of non fastidious organisms.

Recommended for culture of Streptococcus pneumoniae for bile solubility test.

Contains brain and beef heart, peptones, glucose, nacl, buffers.

Also used for preparing inocula for antibiotic sensitivity test

Simple media

- Nutrient media is a basal media.

- Contains peptone water, meat extract - 1%

- addition if 0.5% of glucose makes it a glucose broth.

- addition if 2-3% agar makes it nutrient agar.

- used for simplest and routinely employed medium in laboratory diagnosis.

- if concentration of agar is reduced to 0.2 to 0.4% it becomes semisolid and enables motile bacteria to spread.

Blood Media

Haemolysis can be observed.
