LJ medium

Lowenstein Jensen culture is a growth medium used for mycobacterium especially mycobacterium tuberculosis . Used for diagnosing mycobacterial infections. Also used for testing antibiotic susceptibility and differentiating different species of mycobacteriae

Composition of LJ medium

Malachite green - to prevent growth of other bacteria


Glycerol- helps in growth of mycobacterium tuberculosis


Asparagine - nitrogen coagulated eggs - fatty acids, protein


mineral salt solution.


Low levels of penicillin and nalidixic acid to prevent other bacterial growth.

Culture media for myco. bovis

LJ medium with sodium pyruvate instead of glycerol

LJ Medium color

Green,opaque and opalescent.

Management of LJ medium

Chk medium after 5-7 days of inoculation. Till 8 weeks.


Mycobacterium colonies are colorless dry and rough.
