1. Define micromeritics.
a. Micromeritics – the science and technology of small particles.
i. Particle size affects physical stability and therapeutic response of:
1. Suspensions
2. Emulsions
3. Tablets
4. Capsules
2. Describe the influence of particle size reduction on a dosage form. (describe comminution and the effects of particle size)
a. Comminution – mechanical process of reducing a solid to a smaller state of subdivision.
b. Particle size influences:
i. Appearance
ii. Texture
iii. Taste of drug
iv. Rate of absorption by patient
3. Briefly describe the various methods used to determine/measure a particle size.
a. Sieving: establish dist particle size in powder form, from large holes (top) to progress smaller holes (bottom).
b. Laser Diffraction: shine laser beam on loose particle. Scattered light has pattern unique for size of given material.
c. Scanning Electron Microscope (SEM): probes surface of particle that is covered in a thin layer of gold and bombarded by electrons.
d. Transmission Electron Microscope (TEM): probe internal structure of particle.
5. Briefly explain particle size distribution & importance of surface area.
a. Particle shape affects flow in manufacturing and potentially affect surface area
b. Surface area is important in dissolution rate
c. Absorption Method – determines surface area.
6. Discuss powder characterization & angle of repose. What is the AOR and what is it determined by?
a. When bulk granular materials are poured onto a horizontal surface, a conical pile will form.
b. The Angle of Repose - The internal angle b/w the surface of the pile and the horizontal surface.
c. Angle of Repose determined by:
i. Friction
ii. Cohesion
iii. Shapes of the particles
