In the feline there are 2 types of retics, the punctate and the aggregate. Which of these types do you count?

Aggregate

What is the purpose of performing a reticulocyte count?

Access response of the bone marrow to the anemia

Which species of animal does not have circulating retics?

Horse

How many retics are seen in this picture of dog blood?

How many retics are seen in this picture of dog blood?

3

What stain is used in preparing a reticulocyte count?

New Methlyene Blue, NMB

What would a retic be called under diff-quik stain?

Macrocytic polychromatic rbc

Which retic is considered the younger kind, aggregate or punctate?

Aggregate

How many RBC's should be in a each field when looking for retics?

100 RBC

What is the observed retic count?

The observed retic count is the number of retics per 1000 RBC's

What is the formula for the corrected retic count?

Patient PCV / normal PCV X observed retic

What is the formula for the absolute retic count?

Take the observed retic (before you change it to a percentage) and multiply it times the total RBC count.

What is the procedure for retic staining, incubated/dry?

Procedure: incubate equal parts blood/NMB at room temp for 15 min. Make thick push smear (since diluted and usually anemic). Air dry and evaluate on oil immersion.

Aggregate retics have less than 5 clumps of reticulum. True or False?

False. Aggregate retics have greater than 5 clumps of reticulum.

What is the procedure for NMB wet-prep?

Procedure: Take dry coverslip unstained smear and place bloodside down onto drop of NMB on slide.

Why is the incubated method the preferred method over the wet-prep method?

Incubated method preferred since difficult to distinguish punctate from aggregate, and harder to read.

Punctuate retics have less than 5 clumps of reticulum. True or False?

True

The wet prep method is used when?

A limited amount of blood available.

What kind of retics are these?

What kind of retics are these?

Aggregate

Your dog has a 15% PCV, and you counted 76 retics. What would be the corrected retic count for this animal?

2.5%

What happens when retics are stained with NMB?

When stained with NMB the ribosomes and other organelles of the immature RBC clump into visible granules.

Define a reticulocyte.

A reticulocyte are immature erythrocytes that contain organelles that are lost as the cells mature. These organelles account for the diffuse blue-gray or polychromatophilic staining of the immature cells with Wright’s stain.

What is considered regenerative in regard to the absolute retic count?

In regard to the absolute rectic count the normal dog and cat the absolute retic count is >60,000/µl. In the anemic animal one would like to see an absolute count of 100,000/µl or higher.

Define reticulocyte count.

A reticulocyte count is an expression of the percentage of RBC's that are reticulocytes.

Feline reticulocytes come in 2 varieties:

punctate and aggregate

Canine reticulocytes are almost always

aggregate

The retic count is performed on oil but a suitable viewing area is found on high power.True or False?

True

Would this be considered a proper area to count your retics?

Would this be considered a proper area to count your retics?

Yes

When counting retics, you view ________ number of fields with ___________ number of retics to get a total count of ________.

10, 100, 1000

In cases of severe blood loss anemia, how long might it take for retics to be seen in blood?

2-4 days

Why might you not see retics in the blood until a few days after an anemic patient is admitted?

The blood may not start regenerating immediately and may take a few days.
