Process where tissue specimen is selected and immersed in Isotonic salt



Note: phase constrast or brightfield microscopy for unstained.


Differential dye for stained


Teasing or dissociation

Process where small pieces of tissue not more than 1mm are placed in slide and compressed with another slide



Squash preparation or crushing

Process of examining sediments or sections where cellular materials are spread over a slude by means of wireloop / applicator



Note : useful for cytological examinations especially cancer diagnosis

Smear preparation (two slude method)

Use of applicator stick or platinum loop where materials applied in zigazag line thruout the slide



Note : avoid too thin and too thick

Streaking

A selected portion of materials is transferred to clean slide and spread moderately thick film by teasing the mucous strands with applicator rick



Note : recommended for fresh sputum,bronchial aspirates, thick mucoid secretions

Spreading

Done by placing a drop of secretion or sediment upon one slide and facing to another clean slide



Note : useful for serous fluid , conc. Sputum, enzymatic lavage samples from gastrointestinal tract and blood smear

Pull apart

Special method for smear preparation. The surface of the clean glass slide



Note: they can eamined the cells without deatroying their actual intercellular relationship

Touch preparation / impresion smear

Tjis metjod is utilze when rapid diagnosis of tissue in question is required



Note : recommended when nervous tisaue and lipids asr demostrate

Frozen section

What is the thickness cut from fresh tissue frozen on microtome with CO2 or Cryostat.

10 to 15 um

What is the required atmosphere to kept in cold chamber

-10 to -20°C

Applications in histochemical for both fixed or unfixed are commonly used for:

* rapid pathologic diagnosis during surgery


*diagnostic and research enzyme histochemistry


*diagnostic and research demonstation of soluble substances such as lipids and carbohydrates
