Abstract The experiment is conducted in order to identify a randomly given unknown gram-negative microorganism. By comparative analysis of biochemical tests that were performed throughout the semester to find out its characteristics and determine the identification of bacterium out of four potential unknown was given. To figure out the characteristics, T-streak was performed to isolate the sample from broth to a Tryptic Soy Agar plate, along with an additional slant in case something wrong with the original plate that can possibly affect the result. The gram-stain process of colonies then reveal its shape and morphology, using aseptic

Organisms are divided into two domains: prokaryotes and eukaryotes, which further diverted in three other domains consists of Bacteria, Archaea and Eukarya , there are major different between these two where prokaryotes are much simpler internal structures, circular DNA, lack nucleus and any membrane bounded organelles and in general smaller in size. Bacteria and Archaea fall into this category, however despite they sharing same characteristics, Archaea are in fact closely relate to Eukarya than Bacteria. The domain bacteria is diverted with various prokaryotes, all pathogen fall under this category but not all bacteria are pathogenic (Madigan et.al

MR test tube inoculated and incubated for 24 hours, ten to fifteen drops of methyl red indicator then added to the cultured. If it turned red then that was the positive for the presence of mixed acids. No color change was negative, however, if the media turned orange, it was inconclusive. VP test tube was inoculate and incubated up to 3 days, vortexed, added Barrits Reagent A (alpha-napthol) and Barrits Reagent B (potassium hydroxide). Red or pink color was positive reaction, no color change was negative, however it could take up to an hour to see the color change. SIM test then performed using inoculating needle in semi-solid media contains casein, amino acids, iron-contain compound and sulfur in form of sodium thiosulfate. Inoculated by stabbing technique and incubated for 24 hours, if the organism could reduce sulfur, a black color would be seen. To test for indole production, four or five drops of Kovak’s reagent was added into the cultured, a red color indicated a positive result. Last was to test for motility , if cloudiness was seen away from the stab line, then motility was

After, then other biochemical tests were performed that were specific to the gram- negative bacteria to start the identification process of the unknown. The results of the gram negative tests lead to the identification of the bacteria Alcaligenes faecalis. The gram stain procedure was the first test performed by isolating a colony from the pure culture streak plate. The gram stain was followed orderly right from the referenced laboratory manual. The test resulted to be Gram- Negative bacteria from the compound microscope which were pink and rod shaped that can be seen with the 100x objective lens with the oil immersion.…

During the phase and process of the bacterial unknown project, vast amounts of test were performed in order to determine an unknown species of bacteria. Within the many procedures of identifying the unknown bacteria, I was first given a tube with the unknown bacteria, from which later on I started subcultures by streaking bacteria from the original tube onto a tryptic soy agar (TSA) slant. Throughout the process of identifying the unknown, new subcultures were produced in order to use the newest bacteria to conduct the tests. Thus, this process allowed the unknown bacteria to grow and maintain. Before a series of test were conducted, first a gram stain test was performed in order to determine the unknown’s gram reaction and its cell morphology.…

The Mixed Broth culture contained two unknown bacterial cells and they either Gram-negative or Gram-positive. The procedures were followed as stated from the course laboratory manual by Meramec Community College (1). The first step was figuring out the unknowns was to separate the two bacteria. In order to do this, the Trypticase Soy agar was used. The streaking method was used to spread the bacteria across the Trypticase Soy agar to isolate the bacteria.…

Information about the Bacteria Pseudomonas aeruginosa During microbiology lab, an unknown bacteria culture in liquid broth was assigned to be identified by conducting a series of various tests. Nearly twenty different tests were conducted on the bacteria, but the most important of these was Gram staining test, gelatin stab test, and oxidase test. The results of these three tests allowed for the determination of the bacteria genus and species.…

The purpose of performing physiological tests is to figure out certain characteristics of microorganisms or to reach a conclusion on what an unknown microorganism is. It can be also used to determine what is needed for bacteria to be cultured in or on certain media. Culture media can either be liquid or solid, and they can be used to identify certain characteristics of different bacterial species. This is indicated by the growth or lack of growth in or on certain media. Solid media can also be used to isolate organisms and provide a means to observe their morphology (Wiley 55).…

The focus of this lab was to identify an unknown organism based on its characteristics and the results from each of the tests. There will be various of test to choose from in order to identify the unknown organism, which will eliminate numerous possibilities and narrow it down to one. All the fundamental skills that we have learned and practiced in the lab will be used to perform on our unknown such as aseptic technique, microscopic examination, the use of differential media, and determining if it’s positive or negative. Performing aseptic techniques is the most crucial step that requires the utilizing of transferring, inoculating, and storing bacterial cultures and media. Aseptic technique is defined as procedures that prevent contamination…

Introduction Identifying unknown bacterium is not only vital to microbiology, but also to the medical and pharmaceutical industries in order to treat disease/infection properly. Bacteria are classified and identified to distinguish among strains and to group them by criteria of interest (1). There are several different procedures that can be utilized to identify a bacterium. This includes the oxidation test, gram stain, and catalase test. The purpose of this lab report is to show the understanding of all procedures used to identify the bacterium, and explain how the given results led to the identification of the unknown bacterium.…

To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. (1) A gram stain test was used in determining the shape, size and arrangement of the unknown bacteria (1). The unknown organism is heat fixed onto the slide, in order for the organism to be stained.…

I was given unknown bacteria number 28 and asked to run different testing to identify exactly what this bacterium is. I ran 6 test to determine my unknown. These test includes: Gram Stain, SIM, Sucrose, Citrate, MAC, and EMB. Each one of these test are different and they will give a positive or negative feedback which is how we will know which bacterium I have.…

Gram-positive purple coccus bacteria and gram-negative rod/bacillus were identified using the microscope. The purpose of streaking on the MacConkey is to see if gram negative is lactose fermented or not and on the Mannitol salt agar if gram positive microorganism is able to ferment sugar. There were two types of bacteria that grew on the nutrient agar plates which were…

Identification of unknown organisms is useful in a wide variety of settings; from clinical diagnostics to environmental monitoring (1). Species of bacteria can differ fundamentally in aspects ranging from morphology to metabolism. Exploiting these differences can allow for identification of unknown organisms. Gross morphological characterizations are crude, and better replaced by more accurate and specific tests. At a fundamental level, tests such as the Gram stain divide bacteria in to two broad categories based on the characteristics of their cell wall (2).…
