Penciclovir is a structural analogue of acyclovir, which is also effective against HSV or VZV. Like acyclovir, penciclovir is first converted to its monophosphate derivative by viral thymidine kinase and subsequently to penciclovir triphosphate by cellular kinases (De Clercq and Field, 2006). This activated drug inhibits viral DNA polymerase, thus impairing the capability of the virus to replicate within the host cell.
