The purpose of this experiment was to identify the different absorbance levels and response between normal cells and cancerous cells. Also, the difference between resting or stimulated cells and whether they have a difference in absorbance from each other. Inactivated forms of ERK cells don’t react and bind to the antibody on the ELISA plate, therefore this results in a no colour change on the plate. Whereas activated with added phosphate ERK cells react with the antibody and so antibodies do bind and this causes a colour change on the ELISA plate.
From my results it shows that stimulated cells have a higher absorbance compared to unstimulated cells. This therefore highlights that activated cells have higher energy as they bind to antibodies resulting in a higher absorbance. Stimulated cells is not always a bad thing for instance in HMEC cells stimulated cells have no effect to the body however stimulated SKRB3 cells cause cancer and so more damaging to the body. Unstimulated SKBR3 cells have less energy as they are resting cells and so don’t have a major effect on the body.

The effect that CXCL12 treatment on pERK in SKBR3 cells is that due to the cancer cells being more absorbent it actually allows binding of proteins

The initial start point of ERK is when a signalling molecule binds to the receptor on the cell surface; this causes a change in the cell. ERK proteins communicate by actually adding phosphate groups to nearby proteins. If these proteins become mutated in anyway it can cause cancer. This pathway can become truly damaging to the body if it does become mutated. Inhibitors could be used in order to block the effect of cancer and reduce the ability of the cells growing, This relates to the experiment as AMD3100 reduced the binding of the antibodies and showed less cell
