Unknown Bacterial Culture: Qualitative Analysis

Improved Essays
This experiment focuses on using techniques that were learned over the course of the semester for the qualitative analysis and identification of an unknown bacterial culture. Throughout the course, students were introduced to a working knowledge of basic bacterial laboratory techniques. Students were introduced to the concepts of classification, evolution, and growth of microorganisms, as well as a factual and laboratory knowledge of specific microorganism types. In addition, students should have developed an understanding of the microbial ecology of medical and practical uses for microorganisms, and how they relate to basic biological concepts. It is the intent of the course to provide an opportunity to learn the skills necessary to understand

By comparative analysis of biochemical tests that were performed throughout the semester to find out its characteristics and determine the identification of bacterium out of four potential unknown was given. To figure out the characteristics, T-streak was performed to isolate the sample from broth to a Tryptic Soy Agar plate, along with an additional slant in case something wrong with the original plate that can possibly affect the result.…

The focus of this lab was to identify an unknown organism based on its characteristics and the results from each of the tests. There will be various of test to choose from in order to identify the unknown organism, which will eliminate numerous possibilities and narrow it down to one. All the fundamental skills that we have learned and practiced in the lab will be used to perform on our unknown such as aseptic technique, microscopic examination, the use of differential media, and determining if it’s positive or negative. Performing aseptic techniques is the most crucial step that requires the utilizing of transferring, inoculating, and storing bacterial cultures and media. Aseptic technique is defined as procedures that prevent contamination…

In order for all results to be correct, aseptic technique must be used throughout the whole lab. Aseptic technique is the collective method of working and maintaining pure cultures, which requires awareness of where microbes are (4). Aseptic technique is important to make sure that the unknown bacteria is a pure culture, because a mixed culture could prevent the correct identification. A mixed culture is one that consist of 2 or more organisms (2). If there is more than one organism in the culture, there is no way to identify which one is the unknown given.…

To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. (1) A gram stain test was used in determining the shape, size and arrangement of the unknown bacteria (1). The unknown organism is heat fixed onto the slide, in order for the organism to be stained.…

A dichotomous key and observation can be useful tools in identifying microorganism’s taxonomy as a mold, yeast or bacteria. Microbes can be found everywhere in cold, warm, moist or dry environments and molds, yeast and bacteria each have unique characteristics that set them apart from one another and allow them to survive in these environments. Through macro and microscopic observation, different microbes can be observed through the natural liquid and solid phases and be made into slides to be viewed closely under a microscope. Essentially, a random microorganism can be sampled in a dry environment and be identified through observing unique characteristics that would make the microbe be considered either a mold, yeast or bacterium. Using a nutrient agar plate and cotton-tipped swabs for collecting the microbes, colony growths can be observed in the plate and inoculated onto a slide for further microscopic observation.…

The properties of selective and differential media suggest that differences between the bacterial species could be used in this lab to identify these six species but their effectiveness is unknown. The efficacy of identifying a completely unidentified bacterium in a cost-sensitive manner using these test also has yet to be determined. We hypothesized that applying a combination of selective and differential media with biochemical tests would provide a presumptive identification of the first group of six unknown bacteria. We also hypothesized the identification an unknown member of the Enterobacteriaceae family referred to as unknown #53 could be definitively identified using the API 20E and other tests. To test the first hypothesis, samples of four of the six unknown bacteria were streaked onto plates of Mannitol Salt agar, MacConkey agar, and Eosin Methylene Blue agar to examine their growth on the media.…

The first source of error which is highly likely to have impacted the results is the severe contamination present in seven of the eight plates used to culture the bacteria. The species of said microorganisms are unknown, and thus it is not possible to determine the exact effects of the contaminants. However, a study conducted by the Department of Biology, Indiana University, which investigated the growth of particular bacterium when cultured with other species. The results of the laboratory experiments revealed that many microorganisms can impair the growth of, or eliminate bacteria – effectively acting as a bactericidal or bacteriostatic agent (Hibbing, M. E., Fuqua, C., Parsek, M. R., & Peterson, S. B. 2010). If the contaminant species were capable of this behaviour, it is likely the reason for the major discrepancies and high standard error in the data.…

In this experiment some of the methods learned in class, including aseptic techniques, were utilized to help identify an unknown microbe. For the first part of this experiment, a dichotomous key (attachment 1) was created using the information of possible microbes for the unknown as provided by the instructor. The student was then given an agar slant tube containing the unknown #4 microbe to use throughout the course of the experiment.…
