Ubiquity Lab Report

According to text book and handout Microorganisms, are minute living things that individually are usually too small to be seen with unaided eye. Ubiquity means to be present everywhere. Microbiologist believed that microorganisms are everywhere and have tried to prove it. In this experimenting this lab, we had an experiment on ubiquity. This lab wanted to prove the microbiologist that on this planet has microbes everywhere. To understand more about ubiquity experiment, an important terms need to be explained.
Agar is gelatinous- complex polysaccharide obtained from red algae. An agar plate is a Petri dish that has an agar and nutrients used to culture microorganisms. Also, agar can be expand by adding other nutrients such as glucose and other

In bacteria, cell wall is determined what types of bacteria are. Cell wall of bacteria are called peptidoglycan and composed of complex carbohydrate and protein. Additionally, there are three types of bacteria Gram Positive, Gram Negative and Acid fast. Most bacteria on agar plate will be cream, white, or yellow with circular shape.
Yeast are single celled microorganisms and eukaryotic organisms. They are belongs to the kingdom of Fungi. Typically, they have a round- white shape, but it may appear in football

In a lab experiment, it needs to have two types of controls that is negative control and positive control. These controls will make the end result of experiment without any bias. A positive control is an experimental setup which receive test or treatment, so we are expected what might happens in it. On the other hand, negative control is an experimental set up which is not received any test or treatment, it is being observed its nature state.
Purpose
The purpose of this experiment to identify and differentiate between different types, size, and shape of microorganisms; bacteria, mold, and yeast by using agar to culture and observed these microorganisms and how they can be found on different types of surfaces. And determine weather or microbe are

The hypothesis tested was that if the incubation temperature of Escherichia coli is lower or higher than 37°C, the amount of bacterial growth will decrease. The results of the plates which experienced growth without contamination supported the hypothesis. The plate incubated at 4°C was contaminated, and therefore the E. coli colonies present on the plate, if any, could not be counted. The plate incubated at 23°C had no growth, which supported the hypothesis that if E. coli was incubated at temperatures higher or lower than 37°C, no growth would be present. The bacterium did not begin to have any uncontaminated growth until the incubation temperature was set to 37°C, which had 71 noticeable E. coli colonies.…

During the phase and process of the bacterial unknown project, vast amounts of test were performed in order to determine an unknown species of bacteria. Within the many procedures of identifying the unknown bacteria, I was first given a tube with the unknown bacteria, from which later on I started subcultures by streaking bacteria from the original tube onto a tryptic soy agar (TSA) slant. Throughout the process of identifying the unknown, new subcultures were produced in order to use the newest bacteria to conduct the tests. Thus, this process allowed the unknown bacteria to grow and maintain. Before a series of test were conducted, first a gram stain test was performed in order to determine the unknown’s gram reaction and its cell morphology.…

The goal of this experiment was to identify as well as learn about an unknown species of bacteria along with learning some techniques when it comes to process of elimination in a scientific study. To do this, many different tests were completed to help rule out species that were provided on a sheet of paper listing the possible candidates for the unknown microbe. The Bergey’s Manual of Determinative Bacteriology flow chart was also used to narrow down choices as tests were completed, along with a flowsheet of gram positive and negative microbes provided in Microbiology Class. To begin the process of identifying unknown microbe #7, two isolation streaks of the mystery microbe were made in order to determine whether the microbe likes to grow at 30˚C or 37˚C. It was discovered that the microbe likes to grow more at 37˚C. Next, an observation under the microscope as well as a gram stain were conducted. These tests were conducted in order to identify and understand the shape of the microbe and if it was gram…

In order to do the streaking method, an inoculating loop was sterilized with the Bunsen burner, and put into the unknown specimen then placed onto the TSA. After the plate was streaked it was then incubated at 37 degrees Celsius for 48 hours. The second step was observing the agar after incubation and identifying two different bacteria from the TSA. The third step after observation; was taking out the two bacteria with a sterilized loop and streaking it onto two Mueller Hinton plates. This allowed the separate colonies to grow further for testing.…

This indicate that this peptidoglycan cell wall is much thinner compared to the control we observed from bacillus cereus. Gelatin Hydrolysis test: The purpose of the gelatin hydrolysis test was to determine if our organism had the enzyme gelatinase, to hydrolyze gelatin in the gelatin agar plate that it was initially inoculated into. After incubation and applying TCA it appears that it did not in fact have that partially enzyme. And so it result observed was a opaque cloudy dime sized center, with no visible clearing.…

Determining the cell shapes will help with the process of elimination organisms. Under microscopic examination for my unknown 16 broth, it appears to be a coccus shape and a gram positive. Gram staining can determine cell morphology, size, and arrangement. In a positive gram stain, a primary stain called crystal violet is used to stain the cell. For every new stain, mordant, decolorizing agent, and counterstain, it must be rinsed with distilled water.…

This is where the plain 7mm disc went and this acted as the control of the experiment. Before any of the discs were put on the agar plate, the agar plate was prepared by making a bacterial lawn. In order to do this, a cotton swab was dipped into a tube containing E. Coli and then stirred around to insure that the bacteria was evenly distributed.…

A colony is a organism living together in the same area. In this experiment, we tested how many colonies we will have in two different types of agar plates. One will be our control and treatment. As a control, we used our unwashed hands and a Q-tip to swab our hand from the bottom of the thumb to the opposite side. Once we swabbed our hand, we spread the bacteria from the Q-tip to the plate agar.…

Although bacteria can have different cell structures and or ferment/metabolize different components, they also have specific shape and arrangements that can be helpful when determining a bacterium. The most common shapes are coccus (round), bacillus (rod-shape), and spiral (squiggly line). The shape can be accompanied by a…

Identification of unknown organisms is useful in a wide variety of settings; from clinical diagnostics to environmental monitoring (1). Species of bacteria can differ fundamentally in aspects ranging from morphology to metabolism. Exploiting these differences can allow for identification of unknown organisms. Gross morphological characterizations are crude, and better replaced by more accurate and specific tests. At a fundamental level, tests such as the Gram stain divide bacteria in to two broad categories based on the characteristics of their cell wall (2).…
