Abstract
Experiment 3-14 of the Microbiology: Laboratory Theory & Application lab manual was conducted to utilize the various morphological characteristics of organisms for the identification of unknown broth culture sample #26. Staining techniques including the gram stain and capsule stain were applied to initially describe the sample as a gram-positive staphylococci bacterium. Upon further examination and comparison with the gram-positive control, the microorganism was correctly described as having ovoid-shaped streptococci cell morphology, implicating that sample #26 was Lactococcus lactis. As this microorganism is essential to the food industry and has potential uses in the healthcare industry, proper identification of L. lactis through the staining practices used in this experiment will prove useful for further

The focus of this lab was to identify an unknown organism based on its characteristics and the results from each of the tests. There will be various of test to choose from in order to identify the unknown organism, which will eliminate numerous possibilities and narrow it down to one. All the fundamental skills that we have learned and practiced in the lab will be used to perform on our unknown such as aseptic technique, microscopic examination, the use of differential media, and determining if it’s positive or negative. Performing aseptic techniques is the most crucial step that requires the utilizing of transferring, inoculating, and storing bacterial cultures and media. Aseptic technique is defined as procedures that prevent contamination…

This study was done by applying all of the methods that have been learned so far in the microbiology laboratory class for the identification of an unknown bacterium. The first step I took towards identifying my unknown was by using the given unknown broth and doing a streak plate to get single colonies by shaking the solution before taking my sample as well as using the aseptic technique to…

During the phase and process of the bacterial unknown project, vast amounts of test were performed in order to determine an unknown species of bacteria. Within the many procedures of identifying the unknown bacteria, I was first given a tube with the unknown bacteria, from which later on I started subcultures by streaking bacteria from the original tube onto a tryptic soy agar (TSA) slant. Throughout the process of identifying the unknown, new subcultures were produced in order to use the newest bacteria to conduct the tests. Thus, this process allowed the unknown bacteria to grow and maintain. Before a series of test were conducted, first a gram stain test was performed in order to determine the unknown’s gram reaction and its cell morphology.…

Lactococcus lactis has two subspecies, lactis and cremoris, both of which are essential in manufacture of many varieties of cheese and other fermented milk products. Lactococcus lactis is related to other lactic acid bacteria such as Lactobacillus acidophilus in our intestinal tract and Streptococcus salivarius in the mouth. However, Lactococcus does not normally colonize human tissues and differs from many other lactic acid bacteria in its pH, salt, and temperature tolerances for growth, which are important characteristics relevant to its use as a starter culture in the cheesemaking industry. Lactococcus lactis is vital for manufacturing cheeses such as Cheddar, Colby, cottage cheese, cream cheese, Camembert, Roquefort and Brie, as well as other dairy products like cultured butter, buttermilk, sour cream and kefir. It may also…

The lab instructor issued out a test tube labeled with letter ‘I’, which consisted of two unknown bacteria, Gram-positive or Gram-negative that were streaked from a pure culture. Sterile techniques were followed while performing precise instructions as stated in the referenced Laboratory Manual. Example 1: The first procedure performed was done by isolating a pure culture from the mixture onto a solid Trypticase soy agar (TSA) media. Sterile technique was done by flaming the loop until it turned red to ensure that there were no current bacteria on the loop avoiding contamination followed by rapidly flaming the neck of the test tube to prevent the entry and contamination of unwanted microbes.…

To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. (1) A gram stain test was used in determining the shape, size and arrangement of the unknown bacteria (1). The unknown organism is heat fixed onto the slide, in order for the organism to be stained.…

Introduction Identifying unknown bacterium is not only vital to microbiology, but also to the medical and pharmaceutical industries in order to treat disease/infection properly. Bacteria are classified and identified to distinguish among strains and to group them by criteria of interest (1). There are several different procedures that can be utilized to identify a bacterium. This includes the oxidation test, gram stain, and catalase test. The purpose of this lab report is to show the understanding of all procedures used to identify the bacterium, and explain how the given results led to the identification of the unknown bacterium.…

A given test or media may fall into multiple categories. Selective and differential tests are generally less effective at species identification, and consequently, are normally only capable of presumptive identification (3). This lab uses different types of media that are both selective and differential. Mannitol Salt agar (MSA), which contains high levels of sodium chloride kills most bacteria that are not members of the halotolerant Staphylococcus genus. MSA also contains mannitol, which allows differential identification because an indicator detects acid produced by the fermentation of mannitol (4).…

95% ethanol is used as the decolorizing agent, which will wash the crystal violet from Gram-negative bacteria, and a final counterstain to stain bacteria that are no longer stained as a result of the decolorization with ethanol. In our case, the counterstain is basic fuschin, a pink dye. Following this procedure, the Gram-positive bacteria should remain purple and the Gram-negative bacteria should be the pink color of the counterstain. The staining procedure is visualized in Figure 1.1 below. Figure 1.1 Part 1 involved the preparation of a slide containing the above bacterial species, E. coli, B. subtilis, S. epidermidis, and M. catarrhalis.…

Through the use pre-enrichment media that consists of lactose broth, any microorganisms that are not Salmonella wouldn’t be able to survive because of the drop off in pH. After the Salmonella was allowed to growth during the incubation period, Tetrathionate Brilliant Green Both (TBGB) was used to promote the growth of Salmonella, while inhibiting the growth of other microorganisms, such as Gram-positive bacteria as well as Gram-negative bacilli. After the enrichment period, Brilliant Green Agar (BGA) was used as a selective medium for Salmonella, as microbes that are able to ferment lactose and sucrose will appear with yellow butt and slant, while Salmonella will appear red. Finally, the suspected Salmonella colony will then be inoculate in TSI and LIA agar slants in order to be confirm. TSI slants is a test that is use to distinguish microbes that ferments different types of sugar with…
