Rad8 is a conserved protein, it was previously reported to be sensitive towards both UV and gamma irradiation during G1/S period in Schizosaccharomyces pombe (Doe et al., 1993). It consists of three conserved domains, including E3 ubiquitin ligase motif, SNF2 helicase domain, and a family specific HIRAN domain. Rad8 is required for error-free post-replication repair by contributing to polyubiquitylation of proliferating cell nuclear antigen (PCNA). Up-regulation (12.52-fold) of Rad8 indicated that cold and freeze shock caused DNA damage in the cells, Rad8 was expressed to carry out the repair mechanism in order for the cells to survive. Other DNA damage repair proteins, such as cdc36, and NER were expressed when cells were exposed under cold stress, this observation indicated that rapid cold caused severe damage to the DNA, and required several DNA damage repair proteins to ensure an optimal DNA replication take place within the cells. …show more content…
This proteasome requires ATP hydrolysis for ATP-dependent protein degradation of ubiquitinated substrates and 26S proteasome assembly. According to Stone (2014), 26s proteasome regulates E3 ubiquitin ligases, which is an enzyme involved in the final step in the ubiquitination cascade.
