Welcome and Some notes about this Hand book This booklet is your guide to the next 5 practical sessions. It contains all the methods that you need to complete each of the experiments you will carry out step by step. You must refer to these methods during the sessions as the ability to follow a standard method carefully and accurately is an essential skill that any scientist must learn. 1. A clean, well maintained laboratory coat MUST be worn during the practical class. It should be removed before entering areas where food and drink are purchased and/or consumed. 2. Safety spectacles must be worn at all times. 3. Any cuts and abrasions MUST be covered with a waterproof plaster before entering the laboratory. 4. Whilst in the laboratory you MUST NOT eat, drink, smoke or suck your fingers, pencils etc. 5. If your hands become contaminated wash them immediately with disinfectant followed by soap and water. Any spillages of cultures, infected fluids etc. MUST be reported immediately for treatment with concentrated disinfectant. 6. Avoid producing aerosol sprays of microbial cultures, for example during pipetting. 7. Equipment such as slides, and pipette tips should be discarded into the containers of disinfectant (The pink liquid on the bench) immediately after use. As some organisms remain viable for several hours in disinfectant, these containers should be regarded as contagious as soon as contaminated equipment has been placed in them. 8. Be careful not to burn yourself. Do not lean or reach over a lighted Bunsen burner or pick up bottles of medium fresh from the autoclave. Remember a recently flamed tube or bottle will be hot! Long hair should be tied back. 9. At the end of the practical session cultures and equipment should be placed in the appropriate places as indicated. This
